BioToolomics has developed ViralPolish™ 9000 chromatography media for purification of very large size viruses (>100nm), large exosomes, clearance of nucleic acids (>1 kb) etc. The bead has an inert size-exclusion shell and mixed-mode ligand in the core.

This type of media is ideal for rapid clean-up, either as a first step or as a second step, to purify viral particles. Large nanoparticles, such as viruses, are excluded from the beads. The beads can be packed in a column where viruses will pass through the column bed and collected in the flow through fraction, whilst impurities will be captured within the beads. This affords a very gentle purification process resulting in efficient clean up and high recovery of active virus particles.

A typical purification strategy for virus would be to use ViralPolish® as an initial step, to clean up the sample, prior to binding and elution step, typically from an anion exchange media designed for virus purification such as Q SepFast HighRes Plus.

Alternatively, very dilute samples can be purified by first passing through a Q SepFast HighRes Plus or Q SepFast 6HF Plus anion exchange column to concentrate the sample followed by passing through ViralPolish column to remove residual impurities.

ViralPolish™ 9000A and ViralPolish™ 9000B are good for purifying very large size viruses (>100nm), large exosomes, clearance of large DNAs etc. As a guidance, <900KDa molecules could penetrate the shell if loading sample at high flow velocity (e.g. 1 minute contact time); over 9 million Dalton molecules may penetrate the shell if loading samples slowly. “A” type ligand has more ion-exchange property. “B” type ligand has more hydrophobic property. 

Pre-packed columns for screening and process development use are available at following sizes: 1 ml, 5 ml, 3.8 ml (7x100mm ID x BH), 9.5 ml (11x100mm ID x BH), 20 ml (16x100mm ID x BH), 53 ml (26x100mm ID x BH). All these columns have connections fully compatible to Akta systems. Column hardware has 6 bar (0.6 MPa) pressure rating. See Column Instruction for details of HiSep and Optio column hardware.

This range of chromatography media is developed and supported for production scale chromatography use. Regulatory Support File (RSF) is available to assist process validation and submissions to regulatory authorities.

ViralPolish Selection Kit 8000-10000 is suitable for screening large viruses, exosomes, clearance of large DNA fragments (1kb – 10kb) or other large size nanoparticles. It contains 6 x 1 ml pre-packed columns (1 ml each of ViralPolish 8000A, 8000B, 9000A, 9000B, 10000A and 10000B).
